摘要

目的 观察归脾汤合甘麦大枣汤对CUMS模型小鼠抑郁样行为的改善作用,并基于“脑-肠轴”视角探讨其多靶点机制。方法 采用蔗糖偏好实验、强迫游泳实验及旷场实验评价行为学;ELISA检测血清5-HT、TNF-α、IL-10;16S rRNA测序分析肠道菌群;GC-MS测定粪便短链脂肪酸。结果 GMDZ-GP可显著升高蔗糖偏好率、缩短强迫游泳不动时间、延长中央区停留时间;上调血清5-HT水平,降低TNF-α水平,升高IL-10水平。菌群分析显示,GMDZ-GP恢复Shannon多样性,升高丁酸、丙酸含量。结论 GMDZ-GP通过“菌群-代谢-炎症-神经可塑性”网络整合调控脑-肠轴,发挥多靶点抗抑郁效应。

Abstract

Objective To observe the effects of Gupi decoction and Ganmai Dazao decoction on depression-like behavior in CUMS model mice, and to explore the multi-target mechanism based on the perspective of “brain-gut axis”.
Methods Sucrose preference test, forced swimming test and open field test were used to evaluate behavior. ELISA was used to detect serum 5-HT, TNF-α and IL-10. Gut microbiota was analyzed by 16S rRNA sequencing. Fecal short-chain fatty acids were determined by GC-MS.
Results GMDZ-GP significantly increased sucrose preference rate, shortened the immobility time of forced swimming, and prolonged the time of staying in the central area. It upregulated the serum level of 5-HT, decreased the level of TNF-α and increased the level of IL-10. Microbiota analysis showed that GMDZ-GP restored Shannon diversity and increased the content of butyric acid and propionic acid.
Conclusion   GMDZ-GP integrates the brain-gut axis through the "microbiota-metabolism-inflammation-neuroplasticity" network to exert multi-target antidepressant effect.
