目的 研究头穴透刺联合益智地黄汤(Reh)对脑梗死后痴呆(VD)治疗作用及对磷脂酰肌醇-3羟激酶(PI3K)/蛋白激酶B(AKT/PKB)调节氧化应激损伤和细胞凋亡的影响。方法 将小鼠分为4组:假手术组、VD组、多奈哌齐组、头穴透刺地黄汤组。采用水迷宫实验对学生的学习记忆能力进行评价。检测海马组织中caspase-3活性、MDA含量及SOD酶活性。体外将HT22细胞分为4组:假手术组、VD组、多奈哌齐组、头穴透刺地黄汤组。流式细胞术检测细胞内ROS含量及细胞凋亡。结果 与假手术组相比,VD组小鼠学习记忆能力明显下降。海马内p-AKT蛋白表达及SOD酶活性降低,Caspase-3活性及MDA含量明显升高。头穴透刺地黄汤组VD模型小鼠学习记忆能力明显提高,p-AKT蛋白表达及SOD酶活性上调, Caspase-3活性及MDA含量降低。结论 头穴透刺地黄汤通过调节PI3K/AKT途径来减轻氧化应激,抑制细胞凋亡以改善脑功能。
Objective To study the therapeutic effect of scalp acupuncture combined with Yizhi Reh decoction (Reh) on post-stroke dementia (VD) and its effect on phosphatidylinositol-3-hydroxykinase (PI3K)/protein kinase B (AKT/PKB) regulating oxidative stress injury and cell apoptosis. Methods Mice were divided into 4 groups: sham operation group, VD group, donepezil group, and scalp acupuncture Reh decoction group. The water maze test was used to evaluate the learning and memory ability of students. The activity of caspase-3, the content of MDA and the activity of SOD in hippocampal tissue were detected. HT22 cells were divided into 4 groups in vitro: sham operation group, VD group, donepezil group, and scalp acupuncture Reh decoction group. Flow cytometry was used to detect the content of intracellular ROS and cell apoptosis. Results Compared with the sham operation group, the learning and memory ability of mice in the VD group was significantly decreased. The expression of p-AKT protein and SOD enzyme activity in the hippocampus decreased, while the activity of Caspase-3 and the content of MDA increased significantly. The learning and memory ability of VD model mice in the head-point penetrating Rehmannia decoction group was significantly improved, the expression of p-AKT protein and SOD enzyme activity were upregulated, and the activity of Caspase-3 and the content of MDA decreased. Conclusion Head-point penetrating Rehmannia decoction can reduce oxidative stress and inhibit cell apoptosis to improve brain function by regulating the PI3K/AKT pathway.
