“杜仲-牛膝”治疗骨关节炎的网络药理学研究

摘要

目的 “杜仲-牛膝”治疗骨关节炎的网络药理学研究。方法 TCMSP数据库获取“杜仲-牛膝”的活性成分及靶点,再用GEO芯片数据库分析OA相关靶点,Venny平台交集得到治疗靶点。Cytoscape3.9.1软件绘制“杜仲牛膝活性成分-作用靶点”网络。靶点导入STRING数据库分析PPI互作,导入DAVID进行GO和KEGG富集分析。结果 得到“杜仲-牛膝”48个有效活性成分和药物靶点蛋白211个,OA相关基因883个,共有靶点32个。PPI网络发现JUN、MAPK8、PTGS2、MMP9、EGFR等可能是关键靶点。KEGG分析获得99条通路,其中包括TLR信号通路和TNF信号通路。结论 “杜仲-牛膝”有效成分通过多靶点、多信号通路起协同治疗OA作用。

Abstract

Objective To study the network pharmacology of "Eucommia ulmoides - Achyranthes bidentata" in the treatment of osteoarthritis.
Methods The active components and targets of "Eucommia ulmoides - Achyranthes bidentata" were obtained from the TCMSP database, and the relevant targets of OA were analyzed using the GEO chip database. The Venny platform was intersected to obtain the treatment targets. Cytascape 3.9.1 software draws a network of "active ingredients of Eucommia ulmoides and Achyranthes bidentata - action targets". The target was imported into the STRING database to analyze PPI interactions, and DAVID was imported for GO and KEGG enrichment analysis.
Results 48 effective active components, 211 drug target proteins, and 883 OA related genes were obtained from "Eucommia ulmoides - Achyranthes bidentata", with a total of 32 targets. PPI network discovery JUN, MAPK8, PTGS2, MMP9, EGFR, etc. may be key targets. KEGG analysis obtained 99 pathways, including TLR signaling pathways and TNF signaling pathways.
Conclusion   The effective components of "Eucommia ulmoides Oliv. - Achyranthes bidentata" have synergistic therapeutic effects on OA through multiple targets and multiple signal pathways.
